A fair question, so here is the whole answer.

The three-hour window

After a serving of essential amino acids, muscle protein synthesis rises for roughly 90 to 180 minutes and then returns to baseline. In a 2017 study in older men, adding more leucine inside that window did nothing extra. The authors' own recommendation was to take leucine supplements outside of meals (PubMed). Researchers call this the muscle-full effect.

So a bigger single serving is not the answer. A second, separate serving is.

What twice a day looked like in a trial

In healthy older women, a twice-daily leucine-rich beverage raised integrated daily muscle protein synthesis in both rested and exercised legs, and the authors concluded the habit may help attenuate muscle loss (PubMed). A companion paper found the leucine content of the supplement, not its total protein, drove the response (PubMed).

Why the box says what it says

The directions on the carton say between meals or after exercise. We say twice a day, at least three hours apart. Each serving activates muscle comparable to 20g of protein†. Across the day, two spaced servings mean muscle activation comparable to 40g of protein†, for ten calories.

That is why every supply is counted in servings at two a day: 14 to a box, a week per box. Not because we want to sell you double. Because one bigger stick would not do the job.
